Abstract

摘要

Selective permeability is one of the key features of biological membranes. The permeability for small water soluble molecules is controlled on one hand by physico-chemical properties of the lipid bilayer and on the other hand by channel-forming membrane proteins. To transfer the unique properties of natural or artificial channels into technical application a new field emerged called nanopores. The main driving force in this area has been the application of natural pore-forming proteins to transform information stored chemically in the DNA sequence directly into an electric signal in form of a time-dependent ion current. An obvious extension is to use the intrinsic selectivity of channels to detect proteins, peptides or small molecules. Our group is quantifying the uptake of antibiotics into bacteria. For this we extract channels from the outer cell wall of bacteria, reconstitute them into artificial bilayer and characterizing the permeation of small molecules across them.
